Effects of Ulmus macrocarpa Hance Water Extract on Lipid Metabolism in HepG2 Cells

Abstract
This study examined the effects of Ulmus macrocarpa Hance water extract (UME) on the lipid metabolism in HepG2 cells. Oleic acid (OA)-induced HepG2 cells were treated with UME for 24 h, and the lipid accumulation and lipid metabolism-related genes, such as SREBP1c, SREBP2, ACC1, LXRA, GLUT4, CYP7A1, HMGCR, and ACAT2, were analyzed by Oil Red O staining and quantitative real-time polymerase chain reaction. In addition, the lipid metabolism-related enzymes, such as GPDH and PAP, were analyzed by western blot and enzyme activity assay. Treatment of OA-induced HepG2 cells with UME decreased the level of lipid accumulation in the cells. The UME treatment decreased the expression levels and activities of GPDH and PAP enzyme in the cells. The UME treatment also lowered the triglyceride and total cholesterol levels by increasing the expression of CYP7A1 and decreasing SREBP1c, SREBP2, ACC1, LXRA, GLUT4, HMGCR, and ACAT2 mRNA. These results show that the lipid levels in HepG2 cells were reduced by UME, suggesting that it might be a potent functional food for the prevention of hyperlipidemia.
